Disease Introduction: Osteogenesis Imperfecta (OI)
Type: Musculoskeletal disorder
Symptoms:
Fragile bones that fracture easily. Bone pain and skeletal deformities. Curved spine (scoliosis or kyphosis). Blue tint in the whites of the eyes. Hearing loss, bruising easily. Short stature, loose joints, weak muscles. Dental fragility (dentinogenesis imperfecta).
Cause:
Mutations in the COL1A1 or COL1A2 genes, which encode the α1 and α2 chains of type I collagen — the body’s primary structural protein for bone. These mutations result in defective collagen, reducing bone strength and elasticity.
Treatment:
There is no cure for Osteogenesis Imperfecta, but therapy can improve strength, stability, and quality of life.
Occupational therapy assists with daily activities.
Physical therapy strengthens muscles and supports mobility.
Orthopedic surgery uses metal rods or plates to stabilize fragile or curved bones.
Bisphosphonate medication increases bone density.
Growth hormone therapy may be used in selected cases.
Hearing aids, dental care, and fracture prevention strategies help manage complications.
